Oxytocin (Inj)

Oxytocin is a neuropeptide hormone synthesized in the hypothalamus and released from the posterior pituitary. It is best known for roles in parturition and lactation, but is also studied for effects on social bonding, stress reactivity, and neuroendocrine regulation. Research routes commonly include intranasal administration; injectable oxytocin is used clinically for labor/uterine atony.

Dosing Protocols

— 5 mg Vial —

Reconstitute: Add 3.0 mL bacteriostatic water → ~1.67 mg/mL concentration.
Typical daily range: 100–500 mcg once daily subcutaneous (gradual titration).
Easy measuring: At 1.67 mg/mL, 1 unit = 0.01 mL ≈ 16.7 mcg on a U-100 insulin syringe.
Storage: Lyophilized: freeze at −20 °C or refrigerate at 2–8 °C; reconstituted: refrigerate at 2–8 °C for up to 28–30 days.

Schedule: Daily subcutaneous injections for 8–12 weeks.
Cycle Length: 8–12 weeks; optional extension to 16 weeks.
Goal: Explore oxytocin’s effects on social bonding, stress reduction, metabolic function, and pain modulation.

Frequency: Inject once daily subcutaneously.

Phase Dose Syringe (U-100)
Weeks 1–2 100 mcg 6 units (0.06 mL)
Weeks 3–4 200 mcg 12 units (0.12 mL)
Weeks 5–6 300 mcg 18 units (0.18 mL)
Weeks 7–8 400 mcg 24 units (0.24 mL)
Weeks 9–12 500 mcg 30 units (0.30 mL)

— 10 mg Vial —

Reconstitute: Add 3.0 mL bacteriostatic water → ~3.33 mg/mL concentration.
Typical daily range: 100–500 mcg once daily (gradual titration).
Easy measuring: At 3.33 mg/mL, 1 unit = 0.01 mL ≈ 33.3 mcg on a U-100 insulin syringe.
Storage: Lyophilized: freeze at −20 °C or refrigerate at 2–8 °C; reconstituted: refrigerate at 2–8 °C for up to 28–30 days.

Schedule: Daily subcutaneous injections for 8–12 weeks.
Cycle Length: 8–12 weeks; optional extension to 16 weeks.
Goal: Explore oxytocin’s effects on social bonding, stress reduction, metabolic function, and pain modulation.

Frequency: Inject once daily subcutaneously.

Phase Dose Syringe (U-100)
Weeks 1–2 100 mcg 3 units (0.03 mL)
Weeks 3–4 200 mcg 6 units (0.06 mL)
Weeks 5–6 300 mcg 9 units (0.09 mL)
Weeks 7–8 400 mcg 12 units (0.12 mL)
Weeks 9–12 500 mcg 15 units (0.15 mL)
Benefits
  • Induces uterine contractions (clinical obstetrics)
  • Stimulates milk ejection via myoepithelial contraction
  • Modulates social cognition and bonding behaviors (research)
  • May reduce stress responses via HPA-axis modulation (context-dependent)
  • Tool compound for studying oxytocin receptor signaling
Mechanism of Action
  • Binds oxytocin receptor (OXTR), a Gq/11-coupled GPCR
  • Activates PLC→IP3/DAG leading to increased intracellular Ca2+
  • In uterus and mammary glands, Ca2+-dependent smooth muscle contraction results
  • Central effects involve limbic circuitry modulation; peripheral vs central exposure differs by route
Side Effects
  • Headache, nausea
  • Flushing
  • Changes in blood pressure/heart rate
  • Water intoxication/hyponatremia with prolonged high-dose IV (clinical)
  • Uterine hyperstimulation (pregnancy risk)
  • Injection-site reactions
Side Effect Management
  • Avoid prolonged high-dose exposure; monitor fluids/electrolytes in clinical settings
  • Use conservative dosing and monitor cardiovascular response
  • In reproductive contexts, requires medical supervision
  • Prefer intranasal route for CNS-focused research (protocol-dependent)
Contraindications
  • Pregnancy (unless medically supervised for obstetric indication)
  • Significant cardiovascular disease (caution)
  • Hyponatremia risk states (caution with prolonged dosing)
  • Hypersensitivity
